Novel Vector for Making Transgenic Animals for Genetic Regulation

Description

A transgenesis strategy has been designed to downregulate the expression of a gene of interest in a cell-specific manner. This strategy utilizes a cell-specific promoter and an inducible transcriptional regulation system to direct RNAi expression from a transgene cassette. Advantages of this strategy include cell-specific down-regulation of gene expression, and induction elements to permit temporal and quantitative down-regulation of the gene of interest. This strategy circumvents developmental artifacts associated with conventional gene knock out systems. This transgenesis strategy has been successfully employed in zebrafish, but the cassette is adaptable for genetic regulation in multiple organisms.
